Thanks to Bio.Incubate we could inaugurate our own CNC machine today which will help us to prototype our microfluidic chips and develop our MVP to enable single-molecule protein analysis. 🤖🧬

Cofounders Julian Hengsteler & Til Schlotter, in collaboration with Nako Nakatsuka, Ph.D., published this exciting paper demonstrating their technology's ability to target specific amino-acids on peptides with their aptamer-functionalized interface nanopores. 🔍 Key Breakthroughs: 🐢 Slowing down peptide movement to enhance resolution: By slowing down peptide translocation events, their integrated amino acid-specific DNA aptamers ensure single amino acid resolution is achievable. 🎯 Reducing variability in nanopore size: Their innovative approach addresses the variability in solid-state nanopore size & shape, introducing dynamically tunable pores for enhanced sensing accuracy and reproducibility. 🌐 With the possible integration of serial nanopores and additional amino acid-specific aptamers, full #proteinsequencing could be possible! Proud to share our work many years in the making, enabling peptide differentiation through aptamer-functionalized interface nanopores!🧬🔬 We demonstrate that aptamers targeting specific amino acids (phenylalanine) can bind selectively to peptides with vs. without this moiety. Even when phenylalanine is in the peptide backbone, the aptamer still recognizes unique functional groups of this amino acid! We tackled key challenges of stochastic nanopore sensors. We slowed down fast peptide translocations by integrating amino acid-specific DNA aptamers in the nanopore that bind and retain the molecule. We also controlled aptamer-target interactions using size-tunable interface nanopores to confine the binding event. We also decoupled specific (aptamer recognition) vs. nonspecific (electrostatic and hydrophobic) interactions that occur inside the nanopores through complementary techniques. Understanding the mechanisms make the approach generalizable and we hope that this work contributes towards single-molecule proteomics. With this vision, an awesome start-up company, UNOMR led by Til Schlotter and Julian Hengsteler is in the works - keep an eye out for their exciting innovations!

📸 Recap video of the Selection Event 🎥 A fast-paced evening with food, pitches & awards. On Nov. 9th we hosted the Selection Event at the Kunsthaus Zürich when 12 student teams from ETH Zürich & University of Zurich pitched their idea or technology, competing for entry into the Forge or Startup Tracks of BioIncubate. Then, we worked with expert judges to admit the top teams and now are working on the Build Phase 🔨 The Startup Teams, UNOMR & Fempatch, will continue scaling their startup vision & the Forge Teams will prototype their solution to Lonza's challenge in detecting biological contaminants in biomanufacturing. 🚀 Winner of the Forge Track Grand Prize of CHF 30k will be announced in May 2024 based on the best team from the Forge Track. ➡ Want to stay updated on their progress?
