Choline oxidase has wide application in clinical settings and potential use in enzymatic betaine production, but efficient overexpression has been a challenge. Arjun amplify the choline oxidase gene and clones it into an expression vector, optimizing culture conditions for overexpression of the enzyme. The study provides valuable insights into the development of efficient methods for overexpression of choline oxidase for its wide application in clinical and enzymatic betaine production. In addition, the isolated recombinant choline oxidase was immobilized onto Ni-Sepharose beads and loaded in a rotating bed reactor (RBR), optimizing reaction conditions such as pH and temperature for the reaction of choline to glycine betaine via betaine aldehyde. As the main by-product of the olive oil industry, olive leaves have a significant environmental and economic impact. However, there is an increasing demand for their valorization. In a study, Alexandra V. Chatzikonstantinou , Archontoula Giannakopoulou, Stamatia Spyrou, Yannis Simos, Vassiliki Kontogianni, Dimitrios Peschos, Petros Katapodis, Angeliki Polydera, and Haralambos (Haris) Stamatis developed and applied immobilized enzyme batch bioreactors for the chemo-enzymatic treatment of an aqueous Olea europaea leaf extract rich in oleuropein to produce an extract enriched in hydroxytyrosol and other oleuropein hydrolysis products. The biocatalyst was successfully used in a rotating bed-reactor for the modification of the olive leaf extract, leading to high conversion yields of oleuropein (exceeding 90%).
