We aim to bring revolutionary therapies for protecting and restoring vision in patients with advanced glaucoma a significant step closer to clinical deployment.

    From October 31st to November 3rd, 2023, we held our first EGRET-AAA Workshop in the beautiful city of Groningen. Our network organizes biannual workshops at different locations to share project progress, scientific accomplishments, and provide valuable lectures and training activities for our DCs. These gatherings are crucial for building team spirit and fostering a synergistic, productive network.     EMERGING QUANTITATIVE ASSESMENT OF STRUCTURE AND FUNCTION IN ADVANCED GLAUCOMA Dive into the third research work package of EGRET-AAA, focused on revolutionizing clinical assessment methods for advanced glaucoma. The aim of this work package is to develop innovative tools that accurately evaluate structural and functional changes within the retinal and visual pathways. The overall goal is to better stage glaucoma and spot disease progression early on.  Doctoral Candidates will be refining parametric methods like SAP and OCT, expanding the quantitative assessment spectrum for glaucoma. The WP also incorporates magnetic resonance imaging techniques to comprehensively evaluate visual pathway integrity. Tools such as diffusion-weighted imaging and standard/high-resolution functional MRI will help unveil both structural and functional brain integrity. Stay tuned as we venture further into the realm of advanced glaucoma assessment!💡.

    HEALTHY RETINAL ENVIRONMENT: NEUROPROTECTION AND OXYGEN DELIVERY 👁 The second reserach WP of EGRET-AAA develops on five different projects aiming to gain new insights into the genetic pathways and molecules relevant to neuroprotection and oxygen delivery, to offer both remaining and new ganglion cells a healthy environment. In this WP our DCs will bioinformatics analysis through GWAS studies, explore the ferroptotic pathway through molecular screenings, investigate the roles of glia cells in neuroprotection, delve into the vascular and structural losses that occur in glaucoma and understand new predictive risk factors and prognosis indicators for glaucoma to improve early detection and intervention strategies.  Overall, our expected outcome is to discover ways of protecting RGCs and their axons after repair or regeneration via neuroprotective agents and appropriate oxygenation. We also aim to develop a robust, multimodal outcome measure that can be used as a disease/treatment progression assessment. The results will also be directly relevant to current patients with advanced glaucoma as these techniques can help prevent complete blindness by protecting surviving ganglion cells as well as possible. As such, patients may benefit from these efforts long before stem cell therapy becomes a viable clinical option. Stay tuned for more updates! 💡

    EMERGING THERAPIES IN ADVANCED GLAUCOMA: STEM CELL- AND GENE-THERAPEUTIC APPROACHES The first research work package of EGRET-AAA focuses in advancing experimental treatment and regeneration. One of the greatest challenges in glaucoma is how the current treatment options can only slow disease progression but are still incapable of recovering vision loss. This WP aims to revolutionize glaucoma care and restore vision in patients. These experimental therapies will be able to repair or replace retinal ganglion cells and their optic nerve axons. To this end, our DCs will use innovative stem cell, gene and small molecule therapeutic designs in state-of-the-art in vitro and in vivo models. We will generate stem cell derived retinal organoids to test experimental stem cell therapies and regenerate neural outgrowth in vitro, as well as to investigate relevant pathways and potential gene therapy options. Moreover, we will investigate how guidance and neuroprotective molecules can direct axons along a path, allowing for outgrowth over a long distance towards the visual cortex, and to ultimately restore the damaged optic nerve.
