    Topic of the day: Phenotypic instability 💔 Try as we might, microorganisms don’t always do what we design them to do. In a recent mini-review published by our very own Max Guillaume, titled “Assessing and Reducing Phenotypic Instability in Cyanobacteria,” Max discusses a rather insidious obstacle when it comes to the scale-up process of production fermentation: phenotypic instability. Phenotypic instability is a phenomenon where your production culture gradually stops producing over time, and production cannot be rescued from the same culture. The paper outlines how this can be due to the metabolic burden of production itself! But fear not! Max also discusses how we can use genome-scale modelling and various modelling techniques, precisely what we do here at EV Biotech, to tackle and/or mitigate this challenge. Although this review is primarily focused on cyanobacteria, phenotypic instability has been observed in a variety of different organisms and the underlying principles for solving this puzzle are not inherent to one organism. This paper is also closely related to the work of our colleague Anne Hensen, who is currently focusing on modelling strain degeneration during the scale-up process. Phenotypic instability often goes overlooked at the laboratory scale because it is usually only detectable in continuous or large-scale bioreactors, like those you see at the industrial scale.
