    I'm glad to announce that I have completed a CPD course in Transcriptomics by CBEHx, taught by Dr. Veronica Hughes. This intermediate-level course revisited topics in gene regulation and RNA sequencing while also revealing insights into how transcriptomic profiling can bridge the gap between genomics and proteomics, helping me understand what pathological changes can occur and how this can progress to disease. This course also linked the study of toxicogenomics, in which transcriptomics can reveal drug mechanisms of action, resistant/sensitive genes to drugs, and effective target selection. The course highlighted the different high-throughput screening technologies that help with disease diagnosis and the integration of these into clinical practice. The course accentuated the future prospects of transcriptomics and how scientific advancements can contribute to developing personalised medicines, novel classes of chemotherapeutics, and applications in fields outside human biology. With many other topics also explored, this course helped my understanding of molecular biology/biochemistry. I am so grateful for having this opportunity to study a newly emerging field of omics. #transcriptomics #molecularbiology #biochemistry #pharmacology #immunology #CBEHx

    Gene transcription is regulated at two levels: the transcription machinery and chromatin structure. Transcription factors and RNA polymerase II form the pre-initiation complex (PIC), and after initiation, pause control factors regulate pausing, with elongation factors releasing this pause for continued transcription. Chromatin remodeling and histone modifications, such as acetylation and methylation, further control gene expression by altering nucleosome structure and accessibility. Histone modifiers act as signals to activate or repress transcription, making them crucial for fine-tuning gene regulation. #genetics #genomics #transcriptomics

    Immunosenescence refers to the age-related decline in immune function, particularly affecting the adaptive immune system. One of the key components of this system are T cells, which are classified into CD4+ and CD8+ subpopulations, each playing a distinct role in immune regulation and response. Over time, significant changes occur within these T cell populations. In young individuals, the thymus actively supports T cell development, producing a diverse pool of naïve T cells that can respond to new pathogens. However, with aging, the thymus shrinks, leading to reduced production of naïve T cells and a shift towards memory T cells. These memory T cells can be further classified into central and effector memory cells, with some re-expressing markers like CD45RA. The overall diversity of T cell receptors also declines with age, contributing to decreased immune responsiveness. These changes highlight the challenge of maintaining an effective immune response as we grow older, a state known as immunosenescence.

    Type I Hypersensitivity, also known as the immediate or anaphylactic response, involves several steps. First, during antigen sensitization, the allergen is captured by antigen-presenting cells (APCs) and presented to CD4+ T cells. The secretion of interleukin-4 (IL-4) by CD4+ T cells induces their differentiation into T helper 2 (TH2) cells. B-cells are then activated by IL-4 binding to its receptor, IL-4R, leading to the production of IgE antibodies. These IgE antibodies attach to FcεRI receptors on mast cells and basophils. Upon re-exposure to the allergen, these cells degranulate, releasing substances such as histamine and prostaglandin D2. The released substances cause vasodilation and increased permeability of blood vessels. This process involves two main phases: the initial exposure, where sensitization occurs, and re-exposure, which triggers the hypersensitivity reaction.

    RNA processing involves several post-transcriptional modifications that are essential for the maturation of pre-mRNA into functional mRNA, ready for translation. One critical step in this process is capping, which occurs shortly after transcription begins. The cap, known as the 5’ cap, involves the addition of a modified guanine nucleotide to the 5’ end of the pre-mRNA. This cap protects the nascent mRNA from degradation, aids in ribosome binding during translation, and facilitates the export of the mRNA from the nucleus to the cytoplasm. Capping is an essential modification that ensures the stability and functionality of the mRNA molecule.

    When an action potential reaches the axon terminal, it causes the membrane to depolarize, leading to the opening of voltage-gated calcium (Ca²⁺) channels. The influx of Ca²⁺ ions triggers the release of neurotransmitters from synaptic vesicles into the synaptic cleft. These neurotransmitters then bind to receptors on the target cell, allowing positive ions to flow in, which depolarizes the target cell and makes it more likely to fire an action potential. #neuroscience
