Tabcorp chairman Bruce Akhurst said McLachlan was "one of Australia's leading CEOs and securing Gill is a great vote of confidence for Tabcorp's future".
"Gill has a deep understanding of sport, racing and wagering, combined with significant commercial acumen which was highlighted in the substantial growth of AFL revenues under his leadership," he said.
After nearly 10 years as AFL chief executive, McLachlan departed the role late last year.
During his tenure, he oversaw an expansion of the code to 18 clubs and launched the AFLW competition.
He also secured two of Australia's richest media broadcast agreements.
McLachlan takes over from former Tabcorp chief executive Adam Rytenskild, who stepped down in March following a board investigation into allegations of inappropriate and offensive language.
