🔬 Fascinating study recently published in #Science uncovers a new #critical role of #mechanobiology in driving asthma: 📊 The authors report that bronchoconstriction (rapid #contraction of airway smooth muscle cells) not only represents a primary symptom in asthma attacks but also exacerbates the disease by inducing #mechanical #damage to the epithelial barrier. 🗣 "In this work, we present a #mechanical etiology for asthma ... We find that bronchoconstriction causes #pathological airway epithelial crowding, leading to so much cell extrusion that it #destroys the barrier, resulting in the typical postattack inflammation." ⚕ By inhibiting cell extrusion (blocking mechanosensitive ion channels or #contractility), researchers prevented airway damage and reduced inflammation, suggesting #new #therapeutic approaches to asthma management. ❗ Why is this important? 💪This study reinforces that our bodies are (also) mechanical systems & that #mechanobiology is not just a #consequence but a #driver of many diseases, including asthma, and fibrosis. Understanding and manipulating these mechanical processes can lead to first-in-class therapies. 🚀 In this example, not only can #mechanomodulation (i.e. bronchodilators) rescue acute symptoms of asthma, but it also has the potential to prevent chronic inflammation that feeds into driving continued asthmatic responsiveness. What is disease if not cellular #malfunction? Then what is #efficacy if not #restoration of healthy #function? Our work in #functionfirst discovery of anti-fibrotics using our #FLECS Platform is now published in #SLASDiscovery. Here, rather than "hoping" a molecular readout or genetic association ultimately leads to a functional outcome in diseased #fibroblast mechanobiology as unfortunately is typically done, our work represents the first time this critical cellular function is used as a primary endpoint in a massive screening campaign (up to 15,000 molecules / screening day). We accomplish this with our platform built upon proprietary material science, automation, and computer vision that allows us to generate massive datasets describing cell #mechanobiology – a family of #fundamental cellular functions that were not previously addressable at #scale.
