IMI ConcePTION

We collect information from mothers and their doctors to find out what medicines the mother used during pregnancy. We use data from computer models, cell lines and animal lactation studies to predict how medicines transfer to human milk. We collect information about children from mothers who used medicines during pregnancy, to see if there is any correlation between medicine use and malformations or effects on the development of the child. We also collect and analyse milk and blood from breastfeeding mothers who use particular medicines. We have also built a knowledge bank where women and doctors can find reliable information about how safe a medicine is. Want to know more about our work? There is a huge knowledge gap on medicine safety in pregnancy and breastfeeding. Only 3.7% of available drugs are explicitly labeled as safe and information is often inconsistent and difficult to find and understand. A recent study in the British Journal of Clinical Pharmacology is providing reassurance regarding the use of sertraline during pregnancy and breastfeeding. Sertraline is used to manage several mental health conditions, and it is frequently prescribed during pregnancy and breastfeeding. The IMI ConcePTION project has developed a population pharmacokinetic (popPK) model to describe how the body interacts with the drug during pregnancy and breastfeeding. – Our results are in line with previous studies, and show that infant exposure is low, which is reassuring, as sertraline is widely used during pregnancy and breastfeeding, says Anaëlle Monfort, PhD student at the Université de Montréal’s Faculty of Pharmacy, and one of the authors of the recent publication. Did you know that 9 of 10 women use at least one medicine at some stage during pregnancy? Pregnant women sometimes need to take medicines. Sometimes the reason has nothing to do with pregnancy. Sometimes health problems start in pregnancy and sometimes women take medicines before they know that they are pregnant. Often there is not enough evidence to provide trustworthy information about a particular drug might affect the fetus. A new proof of concept study showcasing how population pharmacokinetic approaches can contribute to prediction of infant exposure to medication via breast milk using clinical samples gathered through sparse sampling (i.e. convenient for breastfeeding persons) as well as to identification of risk factors for higher infant exposure. ✅ Infant exposure to sertraline, influenced by CYP2C19 phenotype and breast milk fat content, remains low, providing reassurance regarding the use of sertraline during breastfeeding. 🙏🏿 Thank you to all our colleagues that have contributed to this important effort for breastfeeding patients.
