    Check out this recent article by Sripad Ram et al., from Pfizer, Inc., published in Nature Portfolio's Scientific Reports. The article presents a rigorous assay validation strategy that was used to evaluate the concordance (i.e., accuracy) and reproducibility (i.e., precision) of biomarker detection with of Ultivue’s InSituPlex® (ISP) panels. The authors quantified the accuracy and precision of four different Ultivue InSituPlex® mIF panels, three human OmniVUE™ 4-plex panels, and one custom murine U-VUE® 4-plex panel in tumor tissue specimens with varying levels of T-cell density. Key highlights include: •InSituPlex® panels are typically accurate where the relative difference in cell proportion between a multiplex image and a single-plex image is less than 20% for a given biomarker. •Signal thresholding-based analysis approaches lead to a complex analysis workflow, where global thresholding often results in high variance and necessitates applying local thresholds for each slide for accurate biomarker detection. •Intra- and inter-run co-efficients of variance (CVs) for all biomarkers in OmniVUE™ and U-VUE® panels were within 10-15%, lower than values typically observed with IHC (~25%). •Researchers evaluated the reproducibility of cell-cell distance estimates. A core component of spatial analysis workflows for critical endpoint evaluation, towards generating biological insights, is consistency in cell-cell distance measurements. Evaluation of the reproducibility in cell-cell distance estimates with InSituPlex® technology showed high intra- and inter-run precision. The article demonstrates how InSituPlex® assays enable robust biomarker detection with high concordance and precision, which is critical for implementing multiplexed immunofluorescence assays in pathology.
