Mass spectrometry (MS) faces challenges in detecting low-abundance proteins due to its restricted dynamic range and ion suppression effects from high-abundance proteins. The Proximity Extension Assay (PEA), on the other hand, offers a sensitive alternative for plasma proteomics, detecting proteins at very low concentrations with less pre-analytical processing. While MS provides a broad proteome view, PEA offers targeted detection of low to medium abundance proteins. Combining both can enhance proteome coverage and accuracy, proving valuable for biomarker discovery in plasma proteomics.
