Projects

  • Study of Genetic Variance between three streptococcus species using Random Amplified Polymorphic DNA method

A four weeks project which helped me gain experience on Bacterial DNA isolation and the use of PCR for Random Amplification Of Polymorphic DNA and comparing the genetics variations between 3 streptococcus species.
Also experience working in aseptic conditions to maintain sterile environment.

  • Development of DNA Mrakers for Three Bacillus Species using Restriction Fragment Length Polymorphism

A four weeks project which helped me gain experience on Bacterial cell cloning, isolation of Bacterial DNA, use of restriction endonucleases for RFLP.

  • Modulations in Key Metabolic Pathways of Glucose in Response to Presence of Selected Heavy Metals

    -

    • Studying the effect of three heavy metals of different concentrations on cultured coelomocytes and chloragogen cells of Eudrilus eugeniae (earthworm) and Canna spp. Studied the alterations in central metabolic pathways of glucose i.e. glycolysis, pentose phosphate pathway and Tri carboxylic acid cycle caused by these heavy metals in the cells.

    • Studying the modulations in the activities of three enzymes: hexokinase, glucose-6-phosphate dehydrogenase and malate dehydrogenase representing the above mentioned metabolic pathways. This was achieved by assaying spectrophotometrically for the above three enzymes each.
    • Techniques used: cold-shock treatment for coelomocytes isolation, dissection method for chloragogen cells, primary cell culture, secondary cell culture, trypan blue cell counting method, phase contrast microscopy, spectrophotometer, extraction of enzymes from cells by centrifugation, protein estimation by Folin Ciocalteu method.
