Longitude portfolio company Poseida Therapeutics, Inc. recently announced that the FDA has granted Orphan Drug Designation for the treatment of multiple myeloma to P-BCMA-ALLO1, a novel BCMA-targeted allogeneic, T stem cell memory (TSCM)-rich chimeric antigen receptor (CAR)-T therapy candidate. "The Orphan Drug Designation for P-BCMA-ALLO1 underscores the high unmet medical need for a rapid and accessible off-the-shelf allogeneic CAR-T therapy for patients with multiple myeloma," said Poseida President and CEO Kristin Yarema, Ph.D. "This designation further validates our belief that TSCM-rich allogeneic CAR-T therapies may potentially offer the optimal combination of clinical results, on-demand availability, and high-volume production, while supporting broader access to CAR-T therapies." Streamlining CAR/TCR-T Therapy Manufacturing Chimeric antigen receptor (CAR) T-cell and T-cell receptor (TCR) T-cell therapies are promising treatments for a range of cancers, offering hope to patients whose cancers have become refractory to conventional therapeutic options. Speakers at the recent CAR-TCR Summit Europe in London agreed that despite their efficacy, CAR/TCR-T cell therapy faces challenges that can result in high costs, and limited accessibility. “Investment in advanced therapies is dropping so drug developers are restricting their pipeline development to save costs,” noted Qian Liu, PhD, head of plasmid engineering & lentiviral vectors (LVVs), WuXi Advanced Therapies. “This means many therapies are slower to reach regulatory approval and commercialization and are usually expensively priced, which restricts market and patient access.” https://lnkd.in/e3R4UdGm

An autologous and personalized regulatory T cell therapy is safe in patients with type 1 diabetes, but does not help preserve β-cell function. The investigational personalized autologous cell therapy, dubbed CLBS03, was initially developed by Caladrius Biosciences and consists mainly of the patient’s own regulatory T cells. After harvesting, these cells are allowed to expand in vitro and are functionally enhanced using a proprietary method, which in turn allows them to regulate effector T cells. In 2022, Caladrius merged with Cend Therapeutics and called the resulting entity Lisata Therapeutics. CLBS03's development is also backed by Sanford Health. #biotech #biopharma #T1D #CellTherapy

Yesterday cancer vaccines today in vivo cell therapies (but all mRNA)... Myeloid Therapeutics announced dosing of the first patient in a Ph1 study for MT-303, their second in vivo cell therapy candidate (thanks, Daniel G., for sharing it). Not yet a "crowded" space but its getting there. Avapritinib is the only potent and selective inhibitor approved for the treatment of D842V-mutant gastrointestinal stromal tumors (GIST), the most common primary mutation of the platelet-derived growth factor receptor α (PDGFRA). The approval was based on the NAVIGATOR trial, which revealed overall response rates of more than 90%. Despite this transformational activity, patients eventually progress, mostly due to acquired resistance mutations or following discontinuation due to neuro-cognitive side effects. These patients have no therapeutic alternative and face a dismal prognosis. Notable, little is known about this drug’s binding mode and its medicinal chemistry development, which is instrumental for the development of the next generation of drugs. Against this background, we solve the crystal structures of avapritinib in complex with wild-type and mutant PDGFRA and stem cell factor receptor (KIT), which provide evidence and understanding of inhibitor binding and lead to the identification of a sub-pocket (Gα-pocket).
