Time unstoppably marches on, and so do science and technology!!! With every new development, we find new bottlenecks. When new bottlenecks are overcome by new technology, new heights are achieved, and new possibilities arise! We've all seen it happen. Processes like "cell line development" and "cell line engineering" are relatively nascent, with publications increasing concomitant with recent advances in cell culture, single-cell omics, and gene editing. As the possibilities arise, fields and industries relying on monoclonal cell line development have continued to expand. Attending to security issues around gene synthesis goes beyond just screening for sequences of concern. In this white paper the National Security Commission on Emerging Biotechnology (NSCEB) provides an overview of the broad landscape of security issues and the challenges we need to address. On screening: we don’t yet have concensus for the broader set of sequences that merit concern, a unified screening process, or a robust mechanism to raise alerts when there are concerns. Beyond screening: there is little enforceability and visibility of current synthesis providers, concern about the use of AI to create novel sequences (but hope about the use of AI in countering these concerns!), and a lack of attention to machine tampering and operational security.
