Making the business of CGT commercially viable remains one of the critical challenges for therapy developers. Clinical success is apparent - why aren’t these therapies providing the financial results analysts and drug developers expect? The answer: It all comes down to reimbursement, operations of personalized medicine and cost. The author highlights commercial success for Yescarta and opportunities for other cell therapy providers. The #FDA has granted ADDF-funded APRINOIA Therapeutics a fast track designation for APN-1607, a #PET tracer for imaging #tau protein in patients with progressive supranuclear palsy (PSP). This is a milestone for the field, helping enable #EarlyDetection and accurate diagnosis, which is crucial for treating patients with #neurodegenerative disorders like Alzheimer's and PSP. It will not only serve as the first tau imaging agent for PSP, but will add to the list of available tau imaging tools for #Alzheimers. The development of a vast arsenal of #biomarkers will play an integral role in the administration of #CombinationTherapy and #PrecisionMedicine care regimens that leverage each patient's unique biomarker profile to treat Alzheimer's on an individual basis. 🔬 Exciting News: New Publication on Medrxiv 🌟 Peer review is ongoing, but I wanted to bring it to everyone's attention. 🎉 I'm thrilled to share our latest publication on Medrxiv! This study represents a significant advancement in our understanding of glioblastoma multiforme (GBM) treatment and its outcomes. 🧬 Background: Our research focused on the role of Aldehyde dehydrogenase-1A1 (ALDH1A1), a key enzyme in chemotherapy outcomes, particularly for the standard-of-care treatment, temozolomide, for GBM. Interestingly, we've found a correlation between high ALDH1A1 expression and poor prognosis across various cancers. 🤔 A pivotal discovery was the impact of Proton Pump Inhibitors (PPIs) – commonly prescribed for corticosteroid-induced gastrointestinal issues – as potential inducers of ALDH1A1, potentially affecting patient survival negatively. 📊 Methods: We delved into real-world data from 554 GBM patients, meticulously annotated from electronic medical records (EMR), as part of the XCELSIOR study. Our analysis, using a sophisticated time-varying Cox Proportional Hazard model, took into account various factors such as MGMT methylation status, patient demographics, and PPI use. 📈 Results: The data, spanning 225 cancer centers, revealed a noteworthy trend. Patients on PPIs generally showed a lower median overall survival (mOS) and 2-year survival rates. The effect was most pronounced in patients with MGMT-methylated GBM. 🚨 Conclusions: Our nationwide study strongly indicates the detrimental effects of PPIs on overall survival in GBM patients, especially those benefiting from SOC chemoradiotherapy. I am happy to announce that we have included our 200th evaluable patient in a first randomized-controlled clinical study evaluating a fully Electronic Patient File-integrated Model-Informed Precision Dosing (MIPD) application in kidney transplantation. The MIPD application is compared to physician-directed dosing of tacrolimus, a powerful immunosuppressive drug used as primary agent in de novo renal allograft recipients. After developing a PK learning model that included correlated within-patient variability and after successful in silico testing of the feedback dosing algorithm, a clinical study of 200 individuals (2:1 randomization) was predicted to have sufficient power to demonstrate differences in tacrolimus dosing precision and probability of target attainment (PTA)[CPT Pharmacometrics Syst Pharmacol. 2022 Mar;11(3):348-361 - https://lnkd.in/e4tfGD8x]. This trial was not primarily designed to show differences in early clinical endpoints after renal transplantation, but to assess MIPD performance (versus physicians) on tacrolimus PTA, dosing precision and user-functionality, reliability and safety of the application. Similar tailormade precision dosing algorithms have been developed for testing in lung and heart transplantation. Thanks for the enormous multi-disciplinary team effort to enable the development, set-up, execution and implementation of this study. An amazing team!
