In macromolecular X-ray crystallography, it has long been possible to map particular elements through use of isomorphous replacement or anomalous dispersion methods. This capability has been lacking in cryo-EM. In a cool new approach, to which we made a small contribution, the lab of Bonnie Murphy at the Max Planck Institute has made use of electron energy loss spectroscopy coupled to a workflow for single particle analysis. The method, REEL-EM, has been applied to worm haemoglobin and one of our favorite proteins, the Ryanodine Receptor.
