For several decades, plasmid isolation has been pretty much standard, following resuspension (often includes RNA degradation with RNase), alkaline lysis, neutralization, separation of cell debris, washing, and elution. Moreover, this procedure comes in larger scales of preparation, namely: midiprep, maxiprep, megaprep, and gigaprep. What sets the differences among these scales of preparation (including miniprep) are as follows: 1. Culture volume where plasmids will be extracted 2. Yield Meanwhile, a commendable practice in preparing cultures for plasmid isolation involves overnight 37°C incubation of bacterial culture with a certain speed of orbital shaking. It provides aeration during the incubation period. With aeration in a form of shaking, better growth, better yield. Also, we have to take note of the copy number of the plasmid we are trying to isolate. Definitely, it is convenient to obtain high yields for plasmids with high copy numbers. On the other hand, there are ways to circumvent the challenge in low copy number plasmids. So we have to understand the nature of the plasmids, even the selectable marker (e.g. antibiotic resistance gene/s) they convey. Surely, plasmid isolation is a significant step in cloning as it is our means to acquire vectors in large quantities.
