Yes, you heard it right!! 💥 High Throughput Sequencing (HTS) techniques have been highly employed in the life science field post the discovery of Sanger sequencing. 🔰 What is HTS? 📌 AKA: Massively parallel sequencing 📌 It is the use of automated equipment to rapidly test thousands to millions of samples for biological activity at the model organism, cellular, pathway, or molecular level. 📌Key HTS platform: Illumina, Highseq, Miseq etc., 🔰 Use of HTS in Genomics: ✅Process duration is less, as multiple samples can be sequenced in one go ✅ More effective automated operation - So, contamination and error is minimal ✅ More sensitive and accurate ✅Lower sample volumes are required - So, costs of culture media and reagents can be reduced and much more!! 🔰Scope of HTS: 🔬Gene expression analysis: ChiP Sequencing 💊Epigenetic modification analysis: Methy - seq 🔬Transcriptome analysis: RNA-Seq 💊Protein analysis: Ribo-seq 🔰Steps involved in High Throughput Sequencing: 💠 Extraction: This is the step where you extract the genetic material of interest, RNA or DNA. 💠Enrichment: In this step, enrich the sample you are interested in. Target enrichment involves a subset of genes, which is isolated and sequenced additionally. 💠Quantification: This is where you quantify the enriched material. Depending on the protocol you may need to quantify a control set as well, where you should see no enrichment or only background enrichment.
