We show that CCAR1 acts a splicing modulator of FANCA pre-mRNA and is needed for FANCA protein expression. CCAR1 acts in concert with the U2AF complex to aid in exclusion of a poison exon from the FANCA transcript, ensuring accurate splicing. Loss of CCAR1 leads to inclusion of the poison exon in the FANCA transcript, leading to loss of FANCA protein expression and thereby causing Fanconi Anemia phenotype. #FanconiAnemia #Splicing #RNA #poison_exon
