I had the privilege of representing Beacon at the 2nd Molecular Glue Summit last week and meeting the individuals leading the molecular glue and degrading space. My key takeaways: 🤖 Learning insights about how AI can supplement the forward prediction of molecular glue rational discovery. ❓ Answering questions about ensuring high bioavailability of orally delivered molecular glues. 🤔 How we can comprehensively expand the glueable E3 Ligase landscape beyond CRBN, VHL and DCAF15? 🚀 How to fast-track IND-filing and accelerate the progression of novel molecular glue candidates to the clinic. I was reminded from my conversations over the 3 days that in such a rapidly growing field (58% gross growth last year alone) keeping pace with the preclinical and clinical landscape in depth is unmanageable. As Beacon expands, we're tracking over 200 molecular glues and more than 1,500 degraders at the most granular level of detail to tackle this challenge. Our work "Enhancing Region-Based Geometric Embedding for Gene-Disease Associations" has been accepted and presented in ACM CODS-COMAD 2024, which happened in Bengaluru in Jan 2024. In this work, we tried to use the geometrical knowledge graph embedding (KGE) method to represent biomedical entities as n-dimensional convex-shaped objects in vector space inspired by Kulmanov et al. work(link in paper reference) to perform gene-disease association prediction. We achieved encouraging results. The model is based on the principles of EL++ Description Logic and we used HPO (Human Phenotype Ontology) as our KG/dataset.
