Congratulations to our team member, Dylan Dieters-Castator and his ex-colleagues for the publication of his work as postdoc at Amgen! Dylan and his colleagues optimized a Cell-Surface Capture (CSC) workflow, using magnetic bead-based processing, and through the evaluation of multiple parameters that can affect sensitivity, speed, and cost, including: labeling, enrichment specificity, missed cleavages, nonenzymatic deamidation, data acquisition methods, biosample amounts, and automation, for maximal “Surfaceome” (or “Membranome”, as we call it) coverage and reproducibility. The cell surface proteome represents an important class of molecules for drug and biomarker discovery. However, their enrichment and detection via MS-based proteomics remains challenging due to the low abundance and dynamic nature of the proteins’ presentation at the membrane, post-translational modifications (PTMs), hydrophobic regions, and processing requirements. #drugdiscovery #proteomics https://lnkd.in/epd9MGvH
