Please join the IMAG/MSM WG on Multiscale Modeling and Viral Pandemics and GLIMPRINT, Thursday, June 13th at 3 pm Eastern Daylight Time (EDT) for: Lorenz Adlung, UMC Hamburg-Eppendorf, scMod: Marrying machine learning and deterministic modelling of longitudinal single-cell data Single-cell-based methods such as flow cytometry or single-cell mRNA sequencing (scRNA-seq) allow deep molecular and cellular profiling of biological processes. However, despite their high throughput, these measurements represent only a snapshot in time. But longitudinal single-cell-based datasets can be used for deterministic ordinary differential equation (ODE)-based modeling to mechanistically describe molecular or cellular dynamics. In my talk, I will present two examples of how we are using time-resolved single-cell datasets to gain a better understanding of cellular signaling, immune responses, and tissue regeneration. Our multidisciplinary efforts are focused on developing methods for applying predictive models in biomedical contexts. For example, we envision that deconvolution of time-resolved bulk mRNA sequencing data could complement scRNA-seq resources, e.g. from the Human Cell Atlas, for ODE-based modeling to leverage large-scale single-cell data in clinical practice. And the first publication of my PhD is out! 😊 In this review, we discuss how biomaterials help to better replicate the fibrotic liver microenvironment and present the most relevant in vitro models of human liver fibrosis developed using 3D bioprinting and microfluidics. #Review Advances in Microfluidics for Single Red Blood Cell Analysis by Xiaohao Wang, Xiang Qian and Liwei Lin https://lnkd.in/gAwzpDgd   #MDPI #cell #Microfluidics #medicine #biosensors #sensors #openaccess   #Abstract The utilizations of microfluidic chips for single RBC (red blood cell) studies have attracted great interests in recent years to filter, trap, analyze, and release single erythrocytes for various applications. Researchers in this field have highlighted the vast potential in developing micro devices for industrial and academia usages, including lab-on-a-chip and organ-on-a-chip systems. This article critically reviews the current state-of-the-art and recent advances of microfluidics for single RBC analyses, including integrated sensors and microfluidic platforms for microscopic/tomographic/spectroscopic single RBC analyses, trapping arrays (including bifurcating channels), dielectrophoretic and agglutination/aggregation studies, as well as clinical implications covering cancer, sepsis, prenatal, and Sickle Cell diseases. Microfluidics based RBC microarrays, sorting/counting and trapping techniques (including acoustic, dielectrophoretic, hydrodynamic, magnetic, and optical techniques) are also reviewed. Lastly, organs on chips, multi-organ chips, and drug discovery involving single RBC are described. The limitations and drawbacks of each technology are addressed and future prospects are discussed.

Join Prof Oxana Klementieva from Lund University in this two-part webinar to learn about her recent publication. For the first time, she acquired typical IR spectra and chemical IR images at a sub-micron resolution using the breakthrough technique of O-PTIR. This was achieved without water interference in the protein bands on freshly prepared and hydrated biopsies, tissue slices, and living organisms without compromising the sample's viability and function.   Highlights will include: 💡 The analysis of fresh mouse brain sections on glass slides, imaging beta-sheet-rich amyloid structures. 💡 Time-resolved imaging of living tissues showing the evolution of beta-sheets and lipid oxidation. 💡 A demonstration of how living lung tissue exposed to different levels of IR energy shows no functional impairment. 💡 How the measurement of living salamander embryos, imaged for lipid droplets, remained whole and saw no impact to sample embryo development   Dr Mustafa Kansiz, Director of Product Management at Photothermal Spectroscopy Corp, will provide a brief technique overview, covering submicron simultaneous IR+Raman with co-located fluorescence widefield imaging, with advances now delivering sub-500nm IR spatial resolution.   This two-part event is not just about breakthroughs but about practical breakthroughs. We will showcase a range of application examples from the life sciences, including live-cell lipid metabolism, single-cell bacterial measurements with stable isotope labeling, and fixed tissues for cell typing. Publication announcement 📢   We are excited to share another part of innovative research performed in the NanoBioMedical Centre. Our group of scientists developed two novel hybrid constructs consisting of LLCNPs loaded with hydrophobic MnO-oleate and hydrophilic MnO-DMSA NPs, and further studied in terms of their physico-chemical properties, positive T1 contrast enhancement properties (in vitro and in vivo) and biological safety.   The main reason for the research was to the health risks associated with the use of Gd-chelates and the promising effects of using nanoparticles as T1 contrast agents (CAs) for MRI. Our team decided to focus on Mn-based nanoparticles, which are considered a highly competitive alternative. The use of hybrid constructs with paramagnetic functionality of Mn-based nanoparticles is an effective approach, in particular, the use of biocompatible lipid liquid crystalline nanoparticles (LLCNPs) as a carrier of MnO nanoparticles.   The obtained hybrids differed in terms of particle size-related parameters and colloidal stability, yet both resulted in high relaxivity parameters, in comparison not only to bare MnO NPs but also to commercially developed contrast agents. Our biosafety studies moreover revealed that cell internalization pathways were dependent on the prepared hybrid type, while viability, effects on the mitochondria membrane potential and cytoskeletal networks were rather related to the susceptibility of the particular cell line. The high relaxation rates achieved with the developed hybrid LLCNPs@MnO enable them to be possibly used as novel and biologically safe MRI T1-enhancing CAs in in vivo imaging.   The work performed in the article was supported by the research grant Sonata Bis 6 2016/22/E/ST3/00458 from the NCN National Science Centre, headed by Dr Grzegorz Nowaczyk.

A profound translational gap is one of the major challenges in biomedical research. A unique tissue collection of human coronary arteries and aortae reference tissues allowed for a one-to-one comparison of human and murine atherosclerosis. This analysis clearly defines parallels and incongruences between experimental and clinical atherosclerosis.

🔬 Next milestone for our translational research endeavors 🔬 Soon, the Optica Biophotonics Congress will take place in Fort Lauderdale, Florida, US. I'm looking forward to catch up with colleagues from the field and discuss relevant topics. I am thrilled to be present at this meeting as invited speaker. We begin our highlights of 2023 publications with a focus on our preclinical #MSOT imaging technology (multispectral optoacoustic tomography). Amongst a total of 82, our team has selected the following 2023 preclinical MSOT publication to highlight: "Renal-Clearable Probe with Water Solubility and Photostability for Biomarker-Activatable Detection of Acute Kidney Injuries via NIR-II Fluorescence and Optoacoustic Imaging" In this study, Dr. Shuizu Wu and researchers from her team at the South China University of Technology investigated the use of MSOT to detect kidney function in a model of renal ischemia. Here, optoacoustic signal from a water soluble, renally cleared dye enabled the identification of changes in renal filtration. Microheology has potential for hypercoagulability diagnosis - https://ow.ly/ANcl50PCXLC Research from Biomedical Optics Express [#OPG_BOEx] uses passive microrheology to measure the mechanical properties of clots and obtain structural information with confocal microscopy. As described in their paper “Microrheology and structural quantification of hypercoagulable clots,” the research team successfully observed fibrin clots' structure and microrheology under the same polymerization condition. Their results can encourage a diagnosis test of thrombosis predisposition in patients.
