ABSTRACT

In the present study the Ocimum canum leaf and Platycladus orientalis leaf were selected
for antidiabetic , hepatoprotective and antioxidant activity study.
All the plant parts (Crude drug) were shade dried, powdered, standardized & subjected
for the extraction with water and petroleum ether (40-60 C) in a soxhlet apparatus .
Obtained extracts exhibited the presence of flavonoidal glycosides, tannins,
carbohydrates, terpenoids and sterols constituent in them, when subjected to
phytochemical investigation. It has been observed that the antidiabetic activity exerted by
petroleum ether extract of both plants exhibited maximum percentage . Hence, they were
further screened for invovo antioxidant study . In case of hepatoprotective activity the
aqueous extract of Ocimum canum possess maximum action but in case of Platycladus
orientalis the petroleum ether extract posses maximum action, further its invivo anti
oxidant study was done.

The acute toxicity study for these extracts revealed that selected medicinal plants were
safe up to dose of 2000 mg/kg, hence, 1/10th of this lethal dose i.e. 200 mg/kg b.w. was
taken as effective dose (therapeutic dose). Here both 1/10th i.e- 200 mg/kg and 1/20th
i.e- 100 mg/kg b.w was taken.The acute toxic study was carried according to the OECD
guide line. Here no mortality of any animal species in either sex was observed.

When these extracts i.e. petroleum ether extract of Ocimum canum leaf (200 mg/kg) and
Platycladus orientalis leaf(200 mg/kg) were subjected for antidiabetic activity, it was
confirmed that they possesses marked antihyperglycemic activity by lowering the blood
glucose levels in streptozotocine-induced diabetic rats in single dose (acute) and multi
dose (sub acute) treatment study. The extract treated groups shown prevention of
reduction glucose level in diabetic rats. The hypoglycemic effect comparable to
Glibenclamide (600μg/kg of body weight) suggested that the active fractions may act by
regenerating the β cells in streptozotocine-induced diabetes. After administration of these
extract to rats, it was found that ALT, AST, ALP, CHO, HDL, LDL and VLDL level of
all tested rats were significantly decreased and at the same time PRO level was increased.
Histopathological examination of pancreas of these animals showed comparable
regeneration of Islets of Langerhans and β cells by these extracts and Glibenclamide
standard drug, which were earlier, necroses by streptozotocine.
In order to detect the oxidative stress in diabetic rats its in vivo antioxidant parameter
like reduced glutathione (GSH), thiobarbituric acid reactive substances (TBARS),
Catalase (CAT) and superoxide dismutase (SOD) were determined.This justify the
antioxidant property of both the plant, which also justify the antidiabetic and antioxidant
effect of these plants.
When these extracts screened for hepatoprotective activity, the results obtained from
Paracetamol induced hepatotoxic model in the healthy male Wister rats indicates that,
after the treatment with Paracetamol there was significant rise in SGOT, SGPT, ALP and
total bilirubin levels which were lowered by aqueous extract of Ocimum canum leaf and
petroleum ether extract of Platycladus orientalis leaf, Sylimarin 25 mg/kg was taken as
the standard drug.In the whole process of hepatotoxicity thioacetamide was used for
toxified the liver and it showed same result as like paracetamol intoxification.
In order to detect the oxidative stress in lipid peroxidation in liver i checked it’s in-vivo
antioxidant parameter like reduced glutathione (GSH), thiobarbituric acid reactive
substances (TBARS), Catalase (CAT) and superoxide dismutase (SOD) which justify the
antioxidant property of both the plant. Which also justify the hepatoprotective and
antioxidant effect of the plant.
The biochemical observations were supplemented by histopathological examination of
pancreas and liver section. In histopathological observation it was showed that the
petroleum ether extract of Ocimum canum and Platycladus orientalis possess effectively
antidiabetic and hepatoprotective property.
From this study it was concluded that petroleumether extract of Ocimum canum leaf &
Platycladus orientalis leaf shown antioxidant, hypoglycemic and hepatoprotective action
.
